Acute Toxity Test for Leucozepin™
(Maximum Tolerance Measurement)

Abstract:
Mice were given (ig) Leucozepin™ extract three times daily. The total amount of the compound was 50.01g/kg (equivalent to 457.64g/kg crude drugs), which was 344.42 times the clinical dosage (66g/day) for adults.
Objective:
LD50 can not be measured because of the low toxicity of Leucozepin™; however, its toxicity can be reflected through the measurement of a day’s maximum tolerance.
Materials:
1. Leucozepin™, provided by LIFEnhance, Inc., is a brown extract, with batch number 20020408. 0.11g extract is equal to 1g crude drugs. It is mixed with distilled water into the concentration of 83.33%, and stored in refrigerator at 4°C for use.
2. The Animals, provided by Hubei Animal Experiment Center, were small Kunming-breed mice with the weight of 19±1g, and with the same number of ? (male) ? (female). The mouse food was purchased from the same center.
Methods:
According to the requirement of the literature, 24 small Kunming-breed mice, with the weight of 19±1g, and the same number of ? (male) ? (female), were given nothing but water for 16 hours. They were given 16.67g of the extract at a time (20ml/kg) three times daily and then were observed at least once a day for 7 consecutive days to see their reaction.
Results:
None of the small mice died after taking Leucozepin™ and no abnormal phenomena was observed in their activities except the following: their excrement turned softened and darkened in color. They ate less after being given the extract twice. However, the next day, all activities were normal. According to the formula in the literature, the tolerance times is the total amount of 16.67/Kg medicine ×3=50.01g/kg, being equal to 454.64g/kg of crude drugs.

I.e. The mice tolerance is 344.42 times the adult dosage, which is far beyond 100 times.
